Sepragen developed the SuperFlo® Wedge column in order to simplify method development and the selection of the proper size of a SuperFlo production column. A Wedge column is a cut out version of a  SuperFlo column similar to a pie slice. The Wedge column provides identical bed-height (10, 15 or 20 cm) and surface area ratio of outer and inner frit as a SuperFlo production column. This allows to reliably simulate the flow characteristics of a production column. The Wedge column is packed with a slurry of your column resin similar to a standard SuperFlo column.

A method developed on a Wedge-column can easily be transferred to a production scale SuperFlo column. The developed gradient remains the same on the production column. It is only necessary to adjust the flow rates according to the volume ratio between production and Wedge column. For example: A 100 mL Wedge is used for method development and it should be transferred to a 10 L SuperFlo column. The ratio then is 100 (= 10 L/ 0.1 L). Assuming that the separation on the Wedge is performed at a flow of 30 mL/min, the separation on the 10 L SuperFlo is achieved under the same gradient conditions at 3 L/ min (= 30 mL x 100). SuperFlo Wedge allows the production method development on a small scale, saving time and costs for resin and buffer.
